The read option is going back to the past and taking a traditional game of 11 on 10 and making it 11 on 11....so the answer to your question is neither the 3-4 nor 4-3 would give you an advantage defensing the read option.

What would give you an advantage is a disciplined football team that knows what their assignments are in a read option, and from there, it's one on one football and the best athlete wins.  Defenses started evolving to defense the pass because of the constant fricking rule changes designed to turn the NFL into arena football, but the pendulum has a way of swinging back the other way, so imo, this is what we are seeing, a correction in offensive football as a reaction to take advantage of teams that are built primarily to stop the passing attack.
